Four men aged 23, 23, 24 and 26 died at the scene after a black Seat Cupra left Liverpool Road in Huyton and hit a lamp-post just before 02:00 BST.
A 27-year-old passenger survived and was taken to hospital in stable condition after the car had been travelling from Liverpool toward Knowsley, Merseyside Police said.
Eleven roads, including Liverpool Road and East Prescot Road between Princess Drive and Finch Lane, have been closed for the investigation and are likely to stay shut through the weekend.
Supt Paul Holden said the force's priority is establishing how the crash happened and getting answers for the families of those affected.
